چکیده

It has been recently described that in embryonic stem cells, the expression of some important developmentally regulated genes is repressed, but poised for fast activation under the appropriate stimuli. In this work we show that Bdnf promoters are repressed by Polycomb Complex 2 in mature hippocampal neurons, and basal expression is guaranteed by the coexistence with activating histone marks. Neuronal stimulation triggered by N-methyl-D-aspartate application induces the transcription of these promoters by H3K27Me3 demethylation and H3K27Me3 phosphorylation at Serine 28 leading to displacement of EZH2, the catalytic subunit of Polycomb Repressor Complex 2. Our data show that the fast transient expression of Bdnf promoters II and VI after neuronal stimulation is dependent on acetylation of histone H3K27 by CREB-p/CBP. Thus, regulatory mechanisms established during development seem to remain after differentiation controlling genes induced by different stimuli, as would be the case of early memory genes in mature neurons.

A nitric oxide signaling pathway controls CREB-mediated gene expression in neurons.

Prevailing views of neurotrophin action hold that the transcription factor CREB is constitutively bound to target genes with transcriptional activation occurring via CREB phosphorylation. However, we report that within several CRE-containing genes, CREB is not constitutively bound. Upon exposure of neurons to brain-derived neurotrophic factor (BDNF), CREB becomes rapidly bound to DNA coincident...

Excitatory actions of GABA increase BDNF expression via a MAPK-CREB-dependent mechanism--a positive feedback circuit in developing neurons.

During early neuronal development, GABA functions as an excitatory neurotransmitter, triggering membrane depolarization, action potentials, and the opening of plasma membrane Ca(2+) channels. These excitatory actions of GABA lead to a number of changes in neuronal structure and function. Although the effects of GABA on membrane biophysics during early development have been well documented, litt...

Dietary supplementation with Salvia officinalis L. and aerobic training attenuates memory deficits via the CREB-BDNF pathway in amyloid beta- injected rats

Background: AD is a neurodegenerative disorder in which there is a gathering of beta-amyloid plaques, primarily in the hippocampus, that lead to neuronal death. Exercise training and botanical medications can play a role in the prevention and treatment of neurodegenerative disorders. Objective: The aim of this study was to determine the effects of aerobic training and Salvia officinalis extract...

Brain-derived neurotrophic factor protects rat cerebral cortical neurons from intermittent hypoxia in vitro

Brain-derived neurotrophic factor (BDNF) plays an important role in neuronal survival and growth and participates in neuronal plasticity. BDNF binds to its high affinity receptor TrkB and activates signal transduction cascades crucial for the production of cyclic adenosine mono phosphate (cAMP)-response element binding protein (CREB), which contributes to neurotrophin-mediated survival in the c...

Neurobiology of Disease Low-Level Laser Therapy Rescues Dendrite Atrophy via Upregulating BDNF Expression: Implications for Alzheimer’s Disease

Downregulation of brain-derived neurotrophic factor (BDNF) in the hippocampus occurs early in the progression of Alzheimer’s disease (AD). Since BDNF plays a critical role in neuronal survival and dendrite growth, BDNF upregulation may contribute to rescue dendrite atrophy and cell loss in AD. Low-level laser therapy (LLLT) has been demonstrated to regulate neuronal function both in vitro and i...
